The black-chinned tilapia Sarotherodon melanotheron heudelotii is an ecologically appealing model as it shows exceptional adaptive capacities, especially with regard to salinity. In spite of this, this species is devoid of genomic resources, which impedes the understanding of such remarkable features. De novo assembly of transcript sequences produced by next-generation sequencing technologies offers a rapid approach to obtain expressed gene sequences for non-model organisms. It also facilitates the development of quantitative real-time PCR (qPCR) assays for analysing gene expression under different environmental conditions. Nevertheless, obtaining accurate and reliable qPCR results from such data requires a number of validations prior to interpretation.... |

Eukaryotic chromatin is organized as radial DNA loops with periodical attachments to an underlying nucleoskeleton known as nuclear matrix. This higher order chromatin organization is revealed upon high salt extraction of cells. To understand the sequential change in the functional organization of chromatin during spermatogenesis, we have analysed the higher order organization of chromatin in different testicular cell types and the epididymal sperm of laboratory mouse. The expansion and contraction of the nucleoid DNA following 2 M NaCl extraction was measured in a fluorescence microscope using ethidium bromide (2.5-200 mg/mL) as an intercalating dye to induce DNA positive supercoils. While the halo size varied among cell types (pachytene DNA most extended,... |

Background Considering the similarity between the testis-specific isoform of angiotensin-converting enzyme and the C-terminal catalytic domain of somatic ACE as well as the structural and functional variability of its natural inhibitors, known as bradykinin-potentiating peptides (BPPs), the effects of different synthetic peptides, BPP-10c (<ENWPHQIPP), BPP-11e (<EARPPHPPIPP), BPP-AP (<EARPPHPPIPPAP) and captopril were evaluated in the seminiferous epithelium of male mice. Methods The adult animals received either one of the synthetic peptides or captopril (120 nmol/dose per testis) via injection into the testicular parenchyma. After seven days, the mice were sacrificed, and the testes were collected for histopathological evaluation. Results BPP-10c... |

The large fruit-eating phyllostomid bat, Artibeus lituratus (Olfers, 1818), forearm 69-75 mm, body mass 66-82 g, has a diversified geographic distribution in the Neotropical region. Therefore it is subjected to different climatic conditions that affect its reproduction, leading to different reproductive strategies such as continuous reproduction, seasonal monoestry or seasonal bimodal polyestry. In this study we used morphometric and histological methods to analyze the annual reproductive activity of A. lituratus males in a population living in the Atlantic Forest, Southeastern Brazil. Testis mass, epididymis mass, gonadosomatic index, seminiferous tubule diameter, and Leydig cell nucleus diameter showed no significant differences (p > 0.05) in the two... |

Bisphenol A [2,2-bis(4-hydroxyphenyl)propane] (BPA), 4-nonylphenol (NP) and di(2-ethylhexyl)phthalate (DEHP), and its metabolite mono-2-ethylhexyl phthalate (MEHP) are chemicals found in plastics, which act as endocrine disruptors (EDs) in animals, including human. EDs act like hormones in the endocrine system, and disrupt the physiologic function of endogenous hormones. Most people are exposed to different endocrine disruptors and concern has been raised about their true effect on reproductive organs. In the testis, they seem to preferentially attack developing testis during puberty rather than adult organs. However, the lack of information about the molecular mechanism, and the apparently controversial effect observed in different models has hampered the... |

The spermatogenesis is regulated by the hypotalamus-hypophysis gonadal axis, and the androgens play a fundamental role in their last stages. The administration of the antiandrogen flutamide interferes with the function of androgen dependent organs (Prostate (P) and Seminal Vesicle (VS)). In this study flutamide (10mg/Kg b.w.) was injected to 10 mice and vehicle to the controls (n=6). The mice were sacrificed at 24 (n=5) and 72 (n=5) hours. In testicular sections the tubular diameter (TD) and height of seminiferous epithelium (EH) were measured. P and VS were macerated, and concentrations of fructose (VS) and zinc (P) were determined. No significant difference exists in the DT among the groups. However, the 72 hrs group showed EH higher than the control (p... |

The objectives of this study were to evaluate the effects of pinealectomy, adrenalectomy and pinealectomy-adrenalectomy upon the quantification of spermatogenic cells of rats. As such, 32 adult Wistar rats with a mean body weight of 331.7± 15.5g were assigned into one of the following treatments: (a) a sham-operated control group, consisting of nine animals; (b) ten pinealectomized animals; (c) seven adrenalectomized animals and (d) six pinealectomized plus adrenalectomized animals. No significant differences were observed between groups for the following parameters: body, testes, prostate and seminal vesicle weights, seminiferous tubular diameter, number of cells per seminiferous tubular cross sections (primary spermatocytes at pachytene, round... |

The aim of this study was to evaluate the possible beneficial effects of Catuaba Cristal® (CC), an alcoholic drink made from wine and Erythroxylum catuaba Ar. Cam on testis. Wistar rats either received CC solution (n=8) or water (n=9). Results showed significant body weight reduction within the CC group, although, no weight changes were observed for liver, kidney, testis, epididymis, seminal vesicle and prostate. The volumetric proportion and volume of interstitial tissue and lymphatic space were reduced in the treated group. In the CC group, although the nuclear volume of Leydig cells (LC) decreased, the number of LC per testis increased. These results suggested that CC had no beneficial effect on spermatogenesis of Wistar rats. |

Androgenic anabolic steroids (AAS) are artificial testosterone analogues, used as medicine in chronic diseases, because they increase protein synthesis generating muscle hypertrophy. Its effect has caught the attention of athletes and gym users, thus their consumption has become epidemic, due to easy marketing, the immediate results and the false impression that it doesn't carry health risks. Such risks may globally harm the body. This study aims to investigate the influence on spermatogenesis of using nandrolone decanoate with or without physical training. Twenty-four rats, divided into four groups were used: sedentary group (SG), sedentary on steroids group (SSG), trained group (TG) and trained on steroids group (TSG). The animals were trained on... |

The comprehension of the reproductive cycle allows to understand which are the morphological changes that develop in the gonad during this interval. Thus, many studies have been undertaken in order to describe and classify the stages of gonadal development and reproductive status of Neotropical fishes. For this purpose, specimens of Melanorivulusaff. punctatus were collected in a permanent dam in Chapada dos Guimarães, Mato Grosso, Brazil. The gonads were prepared for analysis by light microscopy. The oogenesis and spermatogenesis have been described, characterizing the stages of gonadal development, together with assessments of the gonadosomatic ratio, germ cell count and verification of variation of mature oocytes in females. Throughout the year the male... |
